The central amygdala (CeA) regulates nociceptive processing and<br \/>\nassociated anxiety in chronic pain. However, the specific microcircuits<br \/>\nand cell types within the CeA that regulate pain-related anxiety have<br \/>\nnot been fully elucidated. In this study, we discovered a microcircuit<br \/>\nin the CeA wherein dynorphinergic neurons of the lateral subdivision of<br \/>\nthe central amygdala (CeL) project to the medial subdivision (CeM),<br \/>\nactivation of which gates anxiogenic effects associated with chronic<br \/>\npain. Dynorphin-mediated activation of CeM astroglial kappa opioid<br \/>\nreceptors (KORs), dependent upon corticotropin-releasing factor (CRF)<br \/>\nreceptor 2 signaling, enhanced CeM neuron excitability by promoting N-<br \/>\nmethyl-D-aspartate receptor (NMDAR) activation, likely via releasing<br \/>\ngliotransmitter D-serine. Behaviorally, KOR-mediated bidirectional<br \/>\ncommunication between CeA peptidergic neurons and astrocytes modulates<br \/>\nthe anxiogenic effect of chronic pain.
